  • Plot: Teenaged science student Miguel O'Hara suffers an accident during a dangerous experiment. He recovers in the hospital and is taken home to his mother Conchata. He later discovers that he has spider-like abilities and realizes he has become the next Spider-Man. His mother is murdered in her own house; the last memory O'Hara has of Conchata is her last words, that he must have gotten his powers "for a reason".
  • Years later, now 25-years-old, O'Hara has since been motivated by his mother's words, and is now an experienced hero. As Spider-Man, he investigates scientist Walker Sloan and discovers that he has been experimenting on Eddie Brock, also known as Venom, but realizes that Brock is not from the same universe. Brock is transformed into Anti-Venom after Sloan deduces that he was being watched. Once Spider-Man learns that Sloan knows he is watching him, all can do is laugh and say "Of course you wisened the --ck up.." before narrowly making his escape.
  • Spider-Man meets his predecessor, Miles Morales, who has since retired and gone into hiding. Realizing that the world is in danger because of Anti-Venom, Morales agrees to help him and dons his Iron Spider suit. They encounter Peter Parker himself as they patrol around New York City for their adversary; Parker advises O'Hara that "With great power comes great responsibility." before disappearing into the night.
  • The due fights Anti-Venom at the park, but they are overpowered and he escapes. They go to the prison to question Alonzo Lincoln, whom O'Hara imprisoned at a younger age. Lincoln reluctantly informs them that Brock was transported to this universe via an unexplainable magic. They confront a regretful Sloan, who agrees to let them use the portal should they be able to save Brock. However, Anti-Venom arrives, brutally eviscerates Sloan, and destroys the portal as he also kills Otto Octavius Jr. He is subsequently given the mechanical arms Octavius Jr. used, and overpowers the spider-men yet again.
  • The duo are confronted by Max Morales, Miles' son, who tries to detain them, but he is stopped by O'Hara's girlfriend Dana D'Angelo, who has been a long-time friend of Max. Realizing the concept of the duo's intentions, Max begrudgingly agrees to help. Anti-Venom attacks Parker Industries, intending to exact revenge on Parker, only for Parker to maintain that he is not in the same universe. He also reveals that he does not care if the world is saved or destroyed, and leaves his employees to die, before the spider-men arrive; Morales is fatally stabbed through his abdomen by Anti-Venom, but not before placing a tracking device on him. Anti-Venom escapes, and Morales dies in Max's arms. A vengeful Max agrees to help Spider-Man find Anti-Venom and stop him.
  • Spider-Man senses that Anti-Venom has kidnapped Dana and goes to rescue her, while Max repairs his father's suit and wears it, thus revealing that he knew he inherited his father's abilities. Spider-Man realizes that Brock cannot be saved, and only heeds half of Parker's insistence on great power, instead following his mother's advice that he became a hero "for a reason", and starts cutting off Anti-Venom's tentacles with Max's help. Spider-Man kills Anti-Venom with his own broken blade and collapses the new multiverse portal the latter made, thus saving not only his world, but the entire multiverse.
  • In the aftermath, Max departs on good terms with Spider-Man, and tells that if he ever needs help, he may ask. Spider-Man replies that Max's father would be proud, before disappearing in the rain. He starts patrolling the city and moonlights around as its silent guardian as the end credits roll.
  • In a post-credits scene, O'Hara sends a message to alternate versions of his predecessors in other universes, indicating that he is offering to help them should they ask; one replies, and it is revealed to be Peter Parker from the universe that caused Brock to be sent to O'Hara's world.
